Your tasks
TSC22D4 is an evolutionarily conserved protein with a long stretch of intrinsically disordered region (IDR) which contains multiple phosphorylation sites. Patients with type 2 diabetes and liver fibrosis have elevated TSC22D4 protein levels in their liver. In this project, you will investigate whether phosphorylation of TSC22D4 contributes to liver fibrosis by performing experiments both in the cell culture and in mouse models. The tasks will involve overexpressing wild type, phospho-defective, and phospho-mimetic TSC22D4 mutants in primary hepatocytes or in mouse livers to address whether TSC22D4 phosphorylation plays role in apoptosis, inflammation and liver injury. The techniques for these measurements will mostly include RNA and protein extraction from the cells or the liver tissue, qPCR analysis, western blotting, ELISA, and immunohistocemical stainings.
Your Profile
- A highly motivated and hardworking student with a Msc degree in biological sciences and a strong academic background in molecular and cellular biology.
- Demonstrated interest and experience in metabolism research including cell culture work and mouse models as well as excellent communication and teamwork skills.
